Magic Red® intracellular cathepsin activity 

in vitro

 

 

ICT’s Magic Red® Assays 는 세포 내 카텝신 활성(intracellular cathepsin activity)을 시험관 내(in vitro)에서 시간에 따라 정량화하고 모니터링할 수 있도록 합니다. Magic Red® detection substrates 는 광안정성 적색 형광체(photostable red fluorophore)인 cresyl violet 을 활용합니다.

 

Cathepsins는 주로 리소좀(lysosomes) 내에 위치하고 활성화되는 유지 단백질분해효소(house-keeping proteases)입니다. 이들은 기질 특이성(substrate specificities)에 따라 정의됩니다. 혈청 또는 세포외 기질 영역(extracellular matrix regions) 내에서 높은 카텝신 효소 활성 수준(elevated cathepsin enzyme activity levels)은 종종 다양한 임상적으로 중요한 병리(clinically significant pathologies)를 나타냅니다. 카텝신 매개 질환(cathepsin-mediated diseases)에는 알츠하이머병(Alzheimer’s disease), 다양한 암(cancers), 그리고 자가면역 관련 질환(autoimmune related diseases)인 관절염(arthritis) 및 골다공증(osteoporosis) 등이 포함됩니다. 골다공증(osteoporosis)은 일반적으로 뼈 구조의 가속화된 분해와 관련이 있습니다.

 

ICT 는 세 가지 서로 다른 선호 특이성(preferred-specificity) Magic Red® 키트를 제공합니다. 각각의 Magic Red® Cathepsin B, Magic Red® Cathepsin K, 및 Magic Red® Cathepsin L detection kit 는 각각의 B, K, 또는 L 카텝신 효소(B, K, or L cathepsin enzymes)를 표적으로 하는 선호 이펩타이드 표적 서열(preferred dipeptide targeting sequences)을 포함하고 있습니다.
