ICT 사의 serine protease inhibitor assays 는 세포를 용해하지 않고 시험관 내(in vitro)에서 세포 내 키모트립신 유사 세린 단백질분해효소 활성(chymotrypsin-like serine protease activity) 을 검출 합니다. 이 시약은 활성화된 세린 단백질분해효소(serine protease enzymes) 에 공유결합하는 비독성의 녹색 또는 적색 형광 억제제(non-cytotoxic green or red fluorescent inhibitors)입니다.

 

이는 시험관 내에서 세포 내 chymotrypsin-like serine protease activity 의 수준을 평가하는 강력한 방법입니다. 세린 단백질분해효소 억제제 프로브(serine protease inhibitor probe)를 세포 배양 배지에 직접 추가하고, 배양한 후 세척 합니다. 이 억제제는 세포 투과성(cell-permeant) 이기 때문에 모든 살아있는 세포 안팎으로 효율적으로 확산됩니다. 세포 내부에 활성화된 키모트립신 유사 효소가 존재하면, 이는 세린 단백질분해효소 억제제와 공유결합하고, 세포 내부에 녹색 또는 적색 형광 신호를 유지합니다.
