소개 (Description)


Glutathione (GSH) is an important tripeptide thiol (-glutamylcysteinylglicine) antioxidant.

 

Glutathione is present in the cells in both the reduced (GSH) and oxidized (GSSG) forms. Under normal physiologic conditions the reduced form (GSH) is predominant due to the action of the NADPH-dependent enzyme GSSG reductase (GR). However, pathophysiologic conditions causing oxidative stress have a profound effect on the cellular thiol balance resulting in a decrease in the GSH/GSSG ratio.

 

BQC GSH/GSSG Assay Kit is a quick, easy, and reproducible assay for determining glutathione and oxidized glutathione in different biological samples.

 

 


분석 원리 (Assay Principles)


BQC GSH/GSSG Assay Kit is based on an enzymatic recycling method that utilizes Ellman′s Reagent (DNTB, 5,5′-dithiobis (2-nitrobenzoic acid)) and glutathione reductase (GR).

 

In the assay, reduced glutathione GSH (native and enzymatically reduced) reacts with DTNB to form a yellow-colored product with a maximum of absorbance at 412 nm. The absorbance measured at 412 nm is therefore directly proportional to the total glutathione concentration (GSH+GSSG) in the sample.

 

This kit can also be used to measure only oxidized (GSSG) by using a specific GSH masking reagent.

 

Reduced glutathione (GSH) in the sample can be determined by subtracting the amount of GSSG from the total amount of glutathione.

 

Glutathione concentration in a sample is determined from a calibration curve using GSSG as standard.
