Premade Single Domain Antibody Libraries


Creative Biolabs currently presents to our worldwide clients multiple well-tailored premade single domain antibody (sdAb or domain antibody) libraries originated from camels, llamas as well as humans. We are happy to leverage our unparalleled library resources and extensive expertise to assist our clients with their sdAb discovery and development projects.


Single domain antibodies, derived from camelid species, represent a special type of antibody consisting of a single monomeric variable antibody domain. With a molecular weight of only 12-15 kDa, single domain antibodies are much smaller than common antibodies (150-160 kDa). Therefore, single domain antibodies combine the benefits of conventional antibodies with important features of small molecule drugs, gaining increasing attention in biological drug development field in recent years. 

 

Advantages of Single Domain Antibodies :

 •   Able to bind to hidden epitopes that are not accessible to whole antibodies

 •   Greater affinity and selectivity, thus promising lower side effect and better efficacy

 •   Penetrate tissues faster than other antibodies, and even break through the blood brain barrier

     oral availability

In addition, we also have two HuSdL® Human Single Domain Antibody Libraries that allow rapid discovery of large numbers of high-potency camelized human single domain antibodies against any therapeutic targets. In contrast with naïve single domain antibody libraries that generate camel-derived antibodies, our library produces camelized human antibodies that have a human origin, allowing for lowest immunogenic potential in humans, especially for long-term and multiple-dose administration.

The two libraries were preselected based on the thermostability and expressibility (in E. coli) of the displayed antibodies. Specifically, in the libraries, the antibody repertoire was heat-treated to remove clones that could not withstand heat-induced aggregation. The genetic codons used to encode the antibody binders are also optimized for bacterial preference. These libraries are invaluable resources for isolating human single domain antibody binders for research, diagnostic and therapeutic applications.


In addition, we are also happy to tailor custom-oriented services of sdAb library construction and screening against a variety of targets.
