Zero-Length Crosslinkers


Zero-length crosslinkers form protein conjugates without adding additional spacer arm atoms between two conjugated molecules. The crosslinker 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC-HCl or EDAC-HCl) is commonly used for peptide synthesis, preparation of immunoconjugates, and protein crosslinking. EDC-HCl carbodiimide is routinely used in concert with N-hydroxysuccinimide (NHS) or N-hydroxysulfosuccinimide (Sulfo-NHS) to enhance EDC-HCl coupling efficiency by acting as a catalyst.

1.  CDI 

 

CDI crosslinker (N,N'-Carbonyldiimidazole) is a zero-length crosslinker that is often employed in the coupling of amino acids for peptide synthesis and also in organic synthesis. CDI can be used to convert alcohols and amines into carbamates, esters, and ureas.

2.  DCC 


DCC crosslinker (N,N'-dicyclohexylcarbodiimide) is a zero-length crosslinker that is often employed in peptide synthesis and organic synthesis. DCC's carbodiimide is useful for coupling amines and carboxylates without the insertion of spacer atoms. DCC crosslinker is commonly used in the synthesis of active ester-containing reagents.


DCC is soluble in a wide range of solvents including: DMSO, DMF, Dichloromethane, Ethyl Acetate, etc. The urea by-product is conveniently insoluble in these solvents, making its removal simple by filtration.

3.  EDC-HCL 


EDC-HCl zero-length crosslinker (EDAC-HCl, EDC or EDAC; EDCI; 1-Ethyl-3-[3-dimethylaminopropyl]carbodiimide hydrochloride) from ProteoChem is ultrapure, and is used to conjugate carboxyl functional groups to primary amines as found in peptides and proteins. It is ideal for covalently binding proteins or peptides to carboxyl containing beads, resins, or other nanoparticals. The convenient 10 mg packaging is great for using ProteoChem's EDC-HCl in Luminex bead conjugation reactions. EDC-HCl is water soluble, thus the reaction can take place at physiological conditions; although the best results take place at pH of 5.5 or less buffered in MES. Sulfo-NHS or NHS is used as a catalyst for EDC-HCl coupling and greatly increases the efficiency of the reaction.

4.  NHS


N-hydroxysuccinimide (NHS) prolongs the lifetime of amino-reactive moites formed at carboxylic acids and increases the efficiency of the zero-length crosslinker EDC. During NHS crosslinking, NHS at concentrtion of 5 mM reacts with EDC to yield a semi-stable NHS ester, that subsequently can be reacted with primary amines (-NH2), resulting in amide crosslinks. NHS is soluble in aqueous and organic solvents, however NHS activation may decrease water solubility of the modified carboxylate molecule. Sulfo-NHS can preserve and even increase the water solubility of targeted molecules, due to the presence of the charged sulfonate moity.

5.  Sulfo-NHS


N-hydroxysulfosuccinimide (Sulfo-NHS) is used in conjunction with EDC-HCl (EDAC-HCl) to convert carboxyl groups to Sulfo-NHS esters which react with primary amines. Sulfo-NHS is used as the catalyst for EDC-HCl coupling and greatly increases the efficiency of the reaction. Sulfo-NHS is ideal for covalently binding protein or peptides to carboxyl containing beads, resins, or other nanoparticals. Sulfo-NHS is water soluble, thus the reaction can take place at physiological conditions.
