PBMCs are separated from peripheral blood by density centrifugation using ficoll, a hydrophilic polysaccharide. The whole blood can be separated into a top layer of plasma, followed by a lower layer of PBMCs, then a fraction of polymorphonuclear cells (such as neutrophils and eosinophils), and finally a bottom layer of erythrocytes. This highly pure population of cells makes them ideal for further isolations of specific cell types within the PBMC fraction.
